ubuntu is built with the gnome desktop and kubuntu is built with the k desktop. As with most distros that offer multiple desktops, you can install one version and then download a different desktop through their package manager. However, it is usually better to install kubuntu if that's what you want than to install ubuntu and then put kubuntu on top of that. For example, changing some settings in one desktop may have unexpected side effects in the other.

1. "KDE in Ubuntu" = Ubuntu base + ubuntu-desktop + kubuntu-desktop
2. "KDE in Kubuntu" = Ubuntu base + kubuntu-desktop
In #1, you basically install kubuntu-desktop (which is Kubuntu's selection of KDE defaults and some of their own customizations) over an existing Ubuntu (GNOME through ubuntu-desktop) installation.
In #2, you get a clean kubuntu-desktop installation.
Other than that. They're exactly the same.
