If you normally use your soundcard - if you have a normal headset, or mic and headphones then get the latest Beta from the Skype website - http://www.skype.com/download/skype/linux/. It used to have problems detecting the soundcard but it is much improved.
If you use USB then DO NOT use the Beta. It made a proper mess of my USB Internet phone. Install the Medibuntu Repository and install Skype from there. It's a slightly older version which works better with USB.
Note that it is a 2 part installation 1) Install Repository 2) Get GPG key
In the past I had to alter my Sound Device Settings to the option with "hw" in it to get it to work. Skype>S Button (bottom left)> OPtions>Sound Deviceds
